In a statement, the works ministry said Fadillah tested positive after a routine Covid-19 test earlier in the morning.
He is in stable condition and will be quarantined at home as he does not have any symptoms.
The ministry said the senior minister for infrastructure previously tested negative on June 3 in his last periodical screening.
“Throughout his quarantine period, Fadillah will continue to ensure that the ministry’s affairs run smoothly,” it said.
The last minister to test positive for Covid-19 was communications and multimedia minister Saifuddin Abdullah, who came down with the virus in April. He has since recovered.
In January, other government leaders had also been infected, including home minister Hamzah Zainudin, economic affairs minister Mustapa Mohamed, women, family and community development minister Rina Harun and national unity minister Halimah Sadique.
